چکیده

Amphiphilic block copolymer micelles have been the focus of research for the last decades because of the promising properties as nano size drug delivery vehicles. In polar aqueous milieu, copolymers self assemble into micelle structures which consist of hydrophilic corona and hydrophobic core regions. Space created inside the micelle core can be used for transportation of drug molecules of hydrophobic nature, low solubility in blood, and with numerous side effects to human body, especially those for cancer treatments. In this paper, I present an overview for use of amphiphilic block copolymers as drug delivery systems by addressing the formation theory of self assembled micelles, factors affecting the efficacy of drug delivery vehicles and a number of methods to improve it. Two main functionalities of drug delivery vehicles with temporal and distribution controls will be discussed. Former term refers to protection of drug molecules from the enzymatic degradation during the circulation in bloodstream and latter refers to accumulation of vehicles around the target site.

Amphiphilic Block Copolymer Nano-micelles: Effect of Length Ratio of the Hydrophilic Block

Block copolymer nano-micelles are especially important in cancer treatment because of their fine dimensions. In this article, three systems of amphiphilic copolymers with similar lengths and different ratios of the hydrophobic and hydrophilic chains are studied using implicit-solvent coarse-grained molecular dynamics simulations. The factor fphil is defined as the ratio of the number...

Doxorubicin-incorporated polymeric micelles composed of dextran-b-poly(DL-lactide-co-glycolide) copolymer

BACKGROUND Polymeric micelles using amphiphilic macromolecules are promising vehicles for antitumor targeting. In this study, we prepared anticancer agent-incorporated polymeric micelles using novel block copolymer. METHODS We synthesized a block copolymer composed of dextran and poly (DL-lactide-co-glycolide) (DexbLG) for antitumor drug delivery. Doxorubicin was selected as the anticancer dr...

Block copolymer micelles: preparation, characterization and application in drug delivery.

Block copolymer micelles are generally formed by the self-assembly of either amphiphilic or oppositely charged copolymers in aqueous medium. The hydrophilic and hydrophobic blocks form the corona and the core of the micelles, respectively. The presence of a nonionic water-soluble shell as well as the scale (10-100 nm) of polymeric micelles are expected to restrict their uptake by the mononuclea...
